Summary of Research: The intermittent exposure of rats to cocaine over several weeks has been shown to produce kindling, a pattern of neuronal facilitation in the brain. Cocaine-induced kindling has been linked to hyperactivity, stereotypic behaviors, and to seizures. A number of anticonvulsant medications, including carbamazepine, phenytoin, and valproate, have been shown to block the development of kindling in animals. It has been speculated that in humans, cocaine-induced kindling may be experienced as intense craving. Anticonvulsant medications given to cocaine abusers may block the development of kindling, thereby eliminating or substantially reducing the craving associated with prolonged cocaine abuse. A number of open-label and double-blind studies are being conducted to examine the efficacy of these anticonvulsants in the treatment of cocaine abuse.
